Résumé : This workshop report describes plans for scientific drilling in the Samail ophiolite in Oman in the context of past, current, and future research. Long-standing plans to study formation and evolution of the Samail crust and upper mantle, involving igneous and metamorphic processes at an oceanic spreading center, have been augmented by recent interest in ongoing, low temperature processes. These include alteration and weathering, and the associated sub-surface biosphere supported by chemical potential energy due to disequilibrium between mantle peridotite and water near the surface. This interest is motivated in part by the possibility of geological carbon capture and storage via engineered, accelerated mineral carbonation in Oman.

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

Mid-ocean ridge basalt (MORB) is the most abundant magma on Earth, and provides a geochemical window into the mantle. Deriving mantle composition and melting processes from the erupted lavas requires correction to be made for their evolution as they pass through and generate the oceanic crust. This is typically done by assuming that modification of melts in crustal magma chambers occurs exclusively by fractional crystallisation. However, extensive mineral major- and trace element data from a full section of fast-spread lower crustal rocks exposed in Hess Deep (equatorial Pacific Ocean) demonstrate that their evolution is instead controlled by reactive porous flow. These reactions lead to a strong enrichment in, and fractionation of, incompatible trace elements in the melt (as recorded by clinopyroxene compositions), leading to melt compositions far outside of the compositional realm of MORB both in terms of trace element abundances and ratios. The reactive signature increases in strength up section, peaking in varitextured gabbros interpreted to represent the fossilised axial melt lens, indicating that reactive porous flow occurred on the scale of the entire lower crust. The enrichment of the melt is coupled with a strong trace element depletion of plagioclase, olivine, and, to a lesser extent, clinopyroxene cores, suggesting that these phases represent the residues of the reactions from which trace elements have been removed. The dominant role of reactive porous flow, and the resulting deviations from fractional crystallisation predictions, suggest that the lower oceanic crust plays a much more complex and significant role in modifying the compositions of MORB than previously expected, with consequent implications for models of mantle processes.

Properly assessing the extent and magnitude of fault ruptures associated with large earthquakes is critical for understanding fault behavior and associated hazard. Submarine faults can trigger tsunamis, whose characteristics are defined by the geometry of seafloor displacement, studied primarily through indirect observations (e.g., seismic event parameters, seismic profiles, shipboard bathymetry, coring) rather than direct ones. Using deep-sea vehicles, we identify for the first time a marker of coseismic slip on a submarine fault plane along the Roseau Fault (Lesser Antilles), and measure its vertical displacement of ∼0.9 m in situ. We also map recent fissuring and faulting of sediments on the hangingwall, along ∼3 km of rupture in close proximity to the fault's base, and document the reactivation of erosion and sedimentation within and downslope of the scarp. These deformation structures were caused by the 2004 M w 6.3 Les Saintes earthquake, which triggered a subsequent tsunami. Their characterization informs estimates of earthquake recurrence on this fault and provides new constraints on the geometry of fault rupture, which is both shorter and displays locally larger coseismic displacements than available model predictions that lack field constraints. This methodology of detailed field observations coupled with near-bottom geophysical surveying can be readily applied to numerous submarine fault systems, and should prove useful in evaluating seismic and tsunamigenic hazard in all geodynamic contexts.

Mid-ocean ridge basalts (MORBs) are the most abundant magmas produced on Earth. They are widely studied to infer mantle compositions and melting processes. However, MORB liquids are also the complex end-product of a variety of intra-crustal processes such as partial or fractional crystallization, melt–rock interaction, and contamination. Deciphering the relative contribution of these different processes is of first-order importance. Contamination at ocean crustal levels is likely, and may occur at magma chamber margins where fresh magmas can interact with previously hydrothermally altered rocks. Characterizing the composition of this crustal contaminant component is critical if we are to understand the relative importance of each component in the resulting MORB liquid.Here we present the results of experiments designed to reproduce the processes occurring at oceanic magma chamber roofs, where crustal contamination should be most extensive, by melting a representative sample of the sheeted dike complex. Anatectic melts thus produced are likely to represent the principal crustal contaminant in MORB. These melts were characterized for major and trace elements, showing B, Zr, Hf, and U enrichment, and Sr, Ti, and V depletion relative to original MORB liquids. In comparison to the starting material, relative element fractionations are observed in the anatectic melts, with enrichments of: U relative to Ba, Nb, and Th; LREE and MREE relative to Sr; and Zr–Hf relative to LREE. Bulk partition coefficients for element partitioning during magma chamber roof anatexis are derived and proposed as valuable tools for tracking MORB contamination.Comparison with natural samples from the East Pacific Rise and the Oman ophiolite shows that anatectic melts can crystallize in situ to form oceanic plagiogranite intrusions, and that residual assemblages associated with the hydrous partial melting stage are represented by hornfelsic dikes and enclaves (also named granoblastic basalts). We now recognize these as commonplace at the root of the sheeted dike complex both at present-day and fossil oceanic spreading centers.
